Offer Description

With the recent changes in western way of life and the continuing decrease in sperm quality, infertility is now a public health issue. Today, the proposed treatments are only palliative and do not attempt to treat the specific defects and bypass several key physiological steps of reproduction. This strategy now reaches its limitations as one in two couple fails to obtain a child after repeated attempts. The goal of this project is to demonstrate that we can use mRNA therapy to cure some genetic cases of male sterility. Several infertile KO mice lines have been created in the lab and the aim of the project is to test new targeted therapies which will permit to restore a functional spermatogenesis in these models. The aim of this PhD will be to restore the defective spermatogenesis present in mouse models of non-obstructive azoospermia (ANO) and severe human oligozoospermia (OS) by nucleotide therapy.

Requirements
Specific Requirements

We are looking for a student with a master’s degree in reproductive biology, motivated and enthusiastic. The PhD student will carry out complex scientific experiments in a reliable and reproducible manner within the allotted time frame need for the project as.

Mice surgery: Injection mRNA into mice testes.
Imagery methods: confocal microscopy, scanning fluorescence microscopy, in vivo imaging, tissue clearing and light sheet microscopy.
Assisted reproductive technologies: in vitro fertilization (IVF) intracytoplasmic sperm injection (ICSI) and sperm cell motility analyses (CASA).
Single sperm cells sorting by flow cytometry and RNAseq analyses.

 

This project involves animal experimentation and a training to obtain the French experimental animal and rodent surgery authorizations will be provided by the lab
