Offer Description
• Carrying out proof of concept of the sensitivity of leukemia cells to drugs targeting the mitochondria based on alterations of the mitochondrial genome
The missions are organized into 3 work packages:
• Creation of a sensitivity matrix (drugs according to mitochondrial alterations) on models of leukemic lines
• Validation of the sensitivity matrix developed on cells from patients suffering from acute myeloid leukemia
• Confirmation of the in vivo response in a mouse model xenografted with leukemia cells from patients of the Precision Medicine strategy for alterations of the mitochondrial genome
• Co-supervision of interns
• Participation in meetings and administrative formalities for collaboration with partners (Optimal, Integragen, etc.)
• Co-drafting of the DAP (Request for authorization to experiment)
• Realization of IC50 by flow cytometry (% mortality, cell cycle)
• Interpretation of results (correlation of mortality data to genomic data, etc.)
• Determination of synergistic activity between 2 molecules via Combenefit software
• Carrying out DNA extraction from cell pellets of leukemic lines or from patient cells
• Cell culture of patient cells and leukemia lines, cell storage
• Animal experimentation on a xenograft mouse model (collaboration with the Optimal animal experimentation platform, injection of treatments, etc.)
• Drafting the application for authorization to experiment and experimental protocols
The Institute for the Advancement of Biosciences (IAB) is an internationally renowned UMR CNRS 5307 INSERM U1209 UGA in fundamental and translational biomedical research based in Grenoble. The IAB was created in 1997. It brings together 300 teacher-researchers, doctors, engineers, doctoral and post-doctoral students as well as technical and administrative staff. The themes focus on epigenetics, clinical research for chronic diseases and cancer. The IAB is located on the UGA health campus, allée des Alpes, 38700 La Tronche. Integration into the Epigenetic Regulations team led by Saadi Khochbin and more particularly in the “pathophysiological and prognostic impact of metabolic and epigenetic deregulations in acute myeloid leukemia (AML)” group led by Professor Mossuz and Dr Mondet which currently includes 1 University Professor-Hospital Practitioner, 1 Associate Professor-Hospital Practitioner, 1 University Hospital Assistant, 2 PhD students, 1 engineer, 1 master intern

Additional Information
Eligibility criteria
Having a PhD in Biology with experience in animal experimentation, flow cytometry and cell culture.
Solid skills in research methods, particularly in the development of protocols, in data analysis and in knowing how to process statistics.
Knowing how to be expected: Communication, Listening, Autonomy, Organization, Rigor, Adaptability
Minimum English level B2, minimum French language B2
