Offer Description
The aim of the project is to understand the molecular bases at the origin of genomic instability of embryonic stem cells. The study will focus on the role of a factor recently identified in the host laboratory involved in this process, its interaction with proteins involved in DNA repair as well as with pluripotency factors in embryonic stem cells. of mice. The candidate will use molecular and cellular biology techniques to study the functional interaction between the DNA repair pathway and pluripotency factors.
-Main tasks
-Cloning and amplification of DNA by PCR
-Protein and chromatin immunoprecipitation from cell lysates
-Expression and purification of recombinant proteins
-Tests of enzymatic activity in vitro
-Immunfluorescence
Secondary tasks:
Trainee supervision (master level)
The Institute of Human Genetics offers an exceptional scientific environment with 22 research teams. One of the major areas of research developed at the institute is genome dynamics and epigenetic control. The main aspects of this research concern DNA replication, maintenance of genome integrity, recombination and meiosis, chromatin and nuclear organization.The project will be developed in collaboration with 4 other groups of the Institute.

Skills
-A very good experience in molecular and cellular biology is required.
-Motivation and passion for science.
-At least one publication of good level (IF> =10) as the first author resulting from the work of thesis.
– Autonomy
-Capacities of organization and communication
– Ability to work in a team
– Knowledge of basic techniques of molecular and cellular biology (western blot, PCR, cloning, immunolabeling).
– Pluripotent stem cells culture
