The Systems Biology and Translational Medicine Laboratory directed by Akhilesh Pandey, M.D., Ph.D. at Mayo Clinic in Rochester, Minnesota, USA, works on basic and translational sciences with the goal of improving and individualizing medicine. We are looking for a motivated and driven molecular biologist with experience in creating and working with in vitro models of genetic disorders including induced pluripotent stem cells (iPSCs), cell models and organoids. Required qualifications include a doctoral degree in the above-mentioned areas with a demonstrated proficiency in tissue culture, generation of induced pluripotent stem cells (iPSCs), differentiation of iPSCs along various lineages and CRISPR-based gene editing. The successful candidate will work with a dynamic and interdisciplinary team of scientists.
Duties and roles
· Work with the PI for hypothesis development and experimental design
· Generate iPSCs from patient-derived primary cells
· Culture and characterize newly generated iPSC lines and differentiate them along various lineages based on needs
· Perform CRISPR-based gene editing and CRISPR-based screens
· Generate organoids from gene-edited cell lines and patient tissues
· Participate in discussions with the research group and presents in conferences/workshops
· Write scientific manuscripts and grant proposals
· Train and work with graduate students and other postdoctoral fellows
Qualification
Education requirements
Doctorate degree is required. Demonstrated significant, original, and creative contributions to a research or creative program or project.
Experience and skills
· Experience with generating iPSCs from primary patient-derived cells
· Experience with differentiation of iPSCs along various lineages such as neuronal cells, osteoblasts, hematopoietic cells, cardiomyocytes etc.
· Willingness to expand the work on additional cell types and systems
· In depth knowledge and experience with CRISPR screens and analysis
· Comfortable working independently and as part of a team
· Ability to track and execute multiple projects at once
· A track record of publications in the above-mentioned areas
· Excellent written and oral communication skills are essential.
About the group
The position to be filled will be in the Systems Biology and Translational Medicine Laboratory at Mayo Clinic, Rochester, Minnesota, USA, led by Dr. Akhilesh Pandey. We are an enthusiastic and multi-disciplinary team of researchers who include mass spectrometrists, molecular biologists, computational biologists, physician-scientists and pathologists. We apply different Omics technologies to solve problems in biology, cancer and genetic diseases. We employ next-generation sequencing technologies including long-read sequencing towards whole genome, exome and transcriptome analysis. We use advanced mass spectrometry platforms (among others) to study proteomics, lipidomics and metabolomics, tailoring our methods to various problems as needed, e.g., to study protein post-translational modifications, protein-protein interactions and perform targeted analyses.
Among the focus areas of the lab are genetic disorders and cancers, with the aim of discovering novel diagnostic and therapeutic targets as well as to carry out functional studies to understand the molecular basis of these disorders. We have been deploying mass spectrometry-based and other methods for multi-omic characterization of genetic disorders including congenital disorders of glycosylation (CDGs), mitochondrial disorders, lysosomal storage disorders and disorders of lipid metabolism. Beyond molecular characterization, we also identify and validate novel biomarkers. We combine discovery and targeted mass spectrometry approaches to develop assays for molecules altered in metabolic disorders, e.g., ceramides, sphingolipids, proteins, peptides and glycopeptides.
